The county was first set up in the state of Virginia on March 17, 1837, when land from Giles and Tazewell counties was taken and used to make it.

The Bluefield, WV-VA Micropolitan Statistical Area has Mercer County as a part of it.

The county has a total area of 421 square miles (1,090 km2), of which 419 square miles (1,090 km2) are land and 1.7 square miles (4.4 km2) (0.4%) are water, according to the US Census Bureau.
