The county is named after Henry Knox, an American Revolutionary War officer who later became the first Secretary of War.

Knox County is a part of the Mount Vernon, OH Micropolitan Statistical Area, which is part of the Columbus-Marion-Zanesville, OH Combined Statistical Area.

The county has a total area of 530 square miles (1,400 km2), of which 525 square miles (1,360 km2) is land and 4.1 square miles (11 km2) (0.8%) is water, according to the United States Census Bureau.
