The county is named for Henry Karnes, a Texas Revolutionary War veteran.
The historic San Antonio and Aransas Pass Railway, which connected San Antonio and Corpus Christi, travelled through Karnes County.
The county has a total area of 754 square miles (1,950 km2), of which 748 square miles (1,940 km2) is land and 6.0 square miles (16 km2) (0.8%) is water, according to the United States Census Bureau.
