Jessamine County is a county in the state of Kentucky in the United States.

As of the census in 2020, there were 52,991 people living there. Nicholasville is the county seat.

In December 1798, the county was made.

The Lexington-Fayette, KY Metropolitan Statistical Area includes Jessamine County.

It is in the Inner Blue Grass area, which has been a center for growing and raising blooded animals, like thoroughbred horses, for a long time.

In the late 18th century, the government set up a business wine industry here.

The US Census Bureau says that the county covers a total area of 175 square miles (450 km2), of which 172 square miles (450 km2) are land and 2.4 square miles (6.2 km2) (1.4% of the total area) are water.

In 2000, farming took up about 129 square miles (330 km2) of the total area of the county.
