Greenup County is a county in the northeastern section of the United States state of Kentucky, along the Ohio River.

The population was 35,962 at the time of the 2020 census.

Christopher Greenup County was established in 1803 and named for him.

Greenup is the county seat.

Greenup County is included in the Huntington-Ashland, WV-KY-OH Combined Statistical Area.

Greenup County, with its northern border created by the Ohio River, was formed on December 12, 1803, by an act of the General Assembly of Kentucky from Mason County, which encompassed the majority of eastern Kentucky at the time.

The county has a total area of 354 square miles (920 km2), of which 344 square miles (890 km2) is land and 10 square miles (26 km2) (2.8%) is water, according to the United States Census Bureau.
